Description:PLGA-PEG-PLGA (hydrogel) (LA:GA 7:1) is a biodegradable thermosensitive hydrogel product that can undergo phase transition at a specific temperature. PLGA-PEG-PLGA (hydrogel) (LA:GA 7:1) can be widely used in drug delivery systems, tissue engineering, wound dressings, and biosensors. The gelation temperature varies with concentration. The higher the concentration, the lower the gelation temperature. The gelation temperature is between 31-34℃ at a concentration of 15-20 wt%.
In Vitro:Gel preparation method: (1) Take the solid raw material out of the freezing environment, return it to room temperature, remove the condensed water on the outer packaging and then open it
(2) According to the gel use environment, add phosphate buffer, complete culture medium or distilled water and other solvents into the bottle to prepare a 15-20% (w/w) solution
(3) Stir at room temperature in a cool environment (≤25°C) for 24 hours until the polymer is completely dissolved to obtain a thermosensitive hydrogel
(4) Cool the prepared thermosensitive hydrogel to 4°C, and then filter and sterilize it with a 0.22 µm polyethersulfone syringe filter under a sterile environment. Note: (1) Generally, the gelling temperature of isotonic solutions such as phosphate buffer and complete culture medium is about 1-2°C lower than that of distilled water
(2) After adding solvent and during dissolution, scrape the gelled solid from the bottle wall, bottle cap, etc. with a scraper or other tools several times to make it fully contact with the solvent, which can accelerate the dissolution of the polymer.
