anti-TNBC agent-13, a NO donor-Aurovertin B conjugate, is a ferroptosis inducer. anti-TNBC agent-13 inhibits GPX4 activity and leads to triple-negative breast cancer (TNBC) cell death. anti-TNBC agent-13 can be used for the research of triple-negative breast cancer[1].

Cell Viability Assay[1]

Cell Line: MDA-MB-231, 4T1, A549
Concentration: 0.625-20 μM
Incubation Time: 48 hours
Result: Showed significant reduction in cell viability in MDA-MB-231 and 4T1 cells, with IC50 values of 1.41 μM and 3.10 μM, respectively. A549 cells showed minimal effect at the same concentration.

Western Blot Analysis[1]

Cell Line: MDA-MB-231
Concentration: 5 μM, 10 μM
Incubation Time: 48 hours
Result: Significantly downregulated GPX4 expression in a concentration-dependent manner, with GPX4/GAPDH ratios decreasing to ~0.25 at 5 μM and ~0.35 at 10 μM.
Cell viability was reduced to ~5% at 100 μM, and the effect was significantly suppressed by Ferrostatin-1 (HY-100579).

(In Vivo)

anti-TNBC agent-13 (compound 7g) (5-10 mg/kg，腹腔注射，每天一次，持续 16 天) 在皮下肿瘤模型中显著抑制 TNBC 进展，并具有良好的安全性[1]。

Animal Model: BALB/c mice (female, 5 weeks old, implanted with 4T1 cells)[1]
Dosage: 5 mg/kg, 10 mg/kg
Administration: i.p.; daily; 16 days
Result: Significantly suppressed tumor growth with a tumor growth inhibition rate of 43.5% at 10 mg/kg. The low-dose (5 mg/kg) group shows a tumor growth inhibition rate of 22.46%.
Maintained stable body weight and showed no signs of distress.
HE staining and Ki-67 immunohistochemistry showed that promoted tumor necrosis and suppressed the proliferation marker Ki-67.
