ULK1 抗体 (YA019)

ULK1 Antibody (YA019) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to ULK1.

ULK1 是一种 serine/threonine-protein kinase，主要参与响应饥饿条件下的自噬过程。它在自噬体前体 autophagophores 的形成中起关键作用，位于 phosphatidylinositol 3-kinase PIK3C3 的上游。ULK1 是自噬调控反馈回路的一部分，既作为 mTORC1 的下游效应因子，又通过与 RPTOR 的相互作用负调控 mTORC1 的活性。它通过 AMPK 的磷酸化被激活，同时通过磷酸化 AMPK 亚基 PRKAA1、PRKAB2 和 PRKAG1 负调控 AMPK 的活性。ULK1 可能在 ATG13/KIAA0652 和 RPTOR 的磷酸化中发挥作用，但需要进一步证据支持。此外，ULK1 在神经元分化的早期阶段发挥作用，是颗粒细胞轴突形成所必需的。它还通过磷酸化 SESN2、SQSTM1、FLCN 和 AMBRA1 来调控自噬过程。其中，磷酸化 AMBRA1 可使其从细胞骨架锚定位点释放，从而诱导自噬体成核。此外，ULK1 通过磷酸化 ATG4B 抑制其蛋白水解激活和去脂化活性，进而抑制自噬。

• 
  Western blot analysis of extracts from MCF-7 (lane 2(20μg), MCF-7 (lane 3(40μg), using ULK1 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.

  Immunohistochemical analysis of paraffin-embedded Mouse brain tissue using ULK1 Antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80366, 1/200) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
