AQP4 (201-220) is an encephalitogenic epitope of AQP-4. AQP4 (201-220) can induce experimental autoimmune encephalomyelitis (EAE) (characterized by midline lesions in the brain, retinal pathology, and lesions at the grey matter/white matter border zone in the spinal cord). AQP-4 is a target antigen in neuromyelitis optica[1].

AQP4 (201-220) 可用于诱导实验性自身免疫性脑脊髓炎 (EAE) 模型[1][2][3][4]。

1. 诱导实验性自身免疫性脑脊髓炎[1][2][3][4]
致病原理
AQP4 (201-220) 通过突破免疫耐受、激活特异性 T 细胞，AQP4 (201-220) 特异性 T 细胞向中枢神经系统 (CNS) AQP4 高表达区域定向迁移，直接杀伤 AQP4+ 星形胶质细胞或激活胶质细胞，引发自身免疫性炎症损伤。
具体造模方法：
小鼠：C57BL6/J, Aqp4⁻/⁻ 小鼠
给药方式：200 μg • 皮下注射 • 单次，注射部位为小鼠尾根部
Note
(1) AQP4 (201-220) 需与含 250/500 μg 结核分枝杆菌 H37Ra 的完全弗氏佐剂 (CFA) 乳化为 200 μL 乳剂给药。
(2) 给药同时静脉注射百日咳毒素 (200 ng/只)，并在第 2 天再给一次。
造模成功指标
临床评分: 0 分：无任何疾病症状；1 分：尾张力丧失 (尾部下垂，无自主摆动)；2 分：翻身能力受损 (无法自主从仰卧位翻转为俯卧位)；3 分：双后肢瘫痪 (后肢无法支撑体重，拖拽行走)；4 分：四肢瘫痪 (前肢+后肢均丧失运动功能)；5 分：濒死状态 (无自主活动，呼吸微弱)。免疫后第 11 天左右开始出现临床症状 (多为 1 分或 2 分)，第 18 天左右达到症状高峰 (评分多为 2-3 分)，第 30 天进入恢复期(评分下降)；若临床评分≥1 分，提示造模成功。
组织学分析: 灰白质交界区出现散在或局灶性炎症病灶；间脑中线区 (第三脑室附近)、第四脑室周围出现炎症病灶；视网膜内层出现炎症浸润，伴随视网膜神经节细胞丢失。
细胞分析: CNS (脑、脊髓、视网膜) 内 CD45+ 免疫细胞和CD4+ T 细胞显著浸润, 病灶区 Iba-1 和 GAFP 阳性细胞数量增多。
分子变化: AQP4 表达降低；IFN-γ, IL-17, CXCL10, CCL20表达升高。
