生物活性

AUH-6-96 is a JAK/STAT signaling inhibitor. AUH-6-96 reduces Unpaired-induced transcriptional activity in Drosophila cells and blocks tyrosine phosphorylation of STAT92E. AUH-6-96 blocks both constitutive and IL-6-induced phosphorylation of STAT3. AUH-6-96 decreases the level of tyrosine-phosphorylated JAK3. AUH-6-96 induces cancer cell Apoptosis by downregulating the expression of anti-apoptotic genes downstream of STAT3. AUH-6-96 selectively reduces the viability of cancer cells with abnormal JAK/STAT signaling pathway. AUH-6-96 is applicable to related research on Hodgkin's lymphoma, breast cancer, and prostate cancer[1].

Western Blot Analysis[1]

Cell Line: Human cancer cell lines (Hodgkin lymphoma L540, breast cancer MDA-MB-468, prostate cancer DU145)
Concentration: 5, 10, 20, 30, 40 μM
Incubation Time: 24 h
Result: Reduced constitutive phospho-STAT3 levels by more than 50% in L540 cells at 10 μM.
Almost completely inhibited phospho-STAT3 levels in L540 cells at 40 μM.
Caused a dramatic reduction in constitutive phospho-STAT3 levels in MDA-MB-468 and DU145 cells at 40 μM.
Left total STAT3 levels unchanged in all cell lines.

Western Blot Analysis[1]

Cell Line: Hodgkin lymphoma L540 cells
Concentration: 5, 10, 20, 30, 40 μM
Incubation Time: 24 h
Result: Almost completely eliminated constitutive phospho-JAK3 levels, while total JAK3 levels remained unchanged.
Inhibited phospho-STAT5 levels in a dose-dependent manner, with total STAT5 levels unchanged.
Did not affect Lyn phosphorylation at 20 μM.
Reduced phospho-Lyn levels but also lowered total Lyn levels at 40 μM.
Caused no changes in phospho-ERK1/2 or total ERK1/2 levels at concentrations up to 40 μM.
