C12FDG (solution) 

C12FDG (5-Dodecanoylaminofluorescein di-β-D-Galactopyranoside) (solution) is a lipophilic green fluorescent substrate for β-galactosidase detection. C12-FDG is more sensitive than Fluorescein di (β-D-galactopyranoside) (HY-101895) for beta-galactosidase activity determinations in animal cells (Ex/Em = 488/523 nm) ^[1].

1. 溶液配置^[2]
1.1 保存储存液
保存：分装后 -20°C 或 -80°C 避光保存，避免反复冻融。
1.2 工作液的配制
用 PBS (或其他缓冲液) 稀释。可根据实际情况稀释相应的储备液。注意，如果溶剂为 DMSO，则必须考虑 DMSO 的细胞毒性，并应制备溶剂对照；如果溶剂为纯水，则工作液在加入细胞前需要过滤并除菌。
注意：工作液现配现用，避光。

2 荧光衰老相关β-半乳糖苷酶 (SA-β-Gal) 实验：
2.1 在 6、12、24 或 96 孔板中以 5×10⁵ 细胞/mL 的密度培养细胞过夜。按照您的常规方案孵育细胞。
2.2 用 200 μL PBS 洗涤细胞一次，然后用 100 μL 固定液 (2% 甲醛/0.2% 戊二醛的蒸馏水) 在室温下固定 5 分钟。
2.3 用 200 μL PBS洗涤细胞2次，用 100 μL 33 μM C12FDG (PBS，pH=6.0) 染色10分钟，再用200 μL Hoechst solution (1 μg/mL Hoechst 33342 (HY-15559) 在 PBS 中，pH 6.0) 10 分钟。
2.4 通过 20× 物镜和 360 nm (Hoechst 33342) 和 480 nm (C12FDG) 激发滤光片对这些细胞进行成像，并分别通过 460 nm 和 535 nm 发射滤光片进行监测。
