CD28-IN-3 is a CD28 inhibitor with an IC50 of 7.80 μM and a Kd of 52.45 μM. CD28-IN-3 selectively blocks the CD28-B7 interaction, thereby inhibiting CD28-mediated costimulation. CD28-IN-3 suppresses the production of proinflammatory cytokines IFN-γ, IL-2 and TNF-α. CD28-IN-3 can be used in the research of checkpoint-resistant cancers[1].

(In Vitro)

CD28-IN-3 (Compound 22VS) (0-100 μM; 2 h) 可在 CHO-K1 细胞中抑制膜结合型 CD28-CD80 与 CD28-CD86 的相互作用，对应的亚微摩尔级 IC50 值分别为 1.48 μM 和 1.39 μM[1]。
CD28-IN-3 (10-300 μM; 24 h) 在浓度高达 300 μM、孵育 24 h 后，对人 Jurkat T 淋巴细胞无细胞毒性[1]。
CD28-IN-3 (0-200 μM; 5 h) 可在 Jurkat-Raji 共培养体系中抑制 CD28 介导的 T 细胞共刺激，其 IC50 为 6.96 μM[1]。
CD28-IN-3 (10-50 μM; 48 h) 以剂量依赖方式在 3D A549 肿瘤-PBMC 共培养体系和人 PBMC-黏膜共培养体系中抑制 CD28 依赖性促炎细胞因子 (IFN-γ、IL-2、TNF-α) 的释放[1]。
