Crystal Violet (solution) 

Crystal Violet (solution) , also known as Gentian violet, methyl violet 10B, is a triphenyl-methane, an alkaline dye that binds to DNA in the nucleus of a cell, staining it a deep purple. It is often used for Gram staining to classify bacteria, or for cell or histological staining^[1].
Solvent and Concentration: Sterile water: 5 mg/mL (0.5%)

使用方法

1.Crystal Violet 工作液的配制
用 20 mL 95% 乙醇溶液溶解 2 g Crystal Violet 与 80 mL 1% 草酸铵水溶液混匀，24 h 后使用。
2.样品处理
2.1 石蜡切片：首先使用二甲苯脱蜡 5-10 min，换用新鲜的二甲苯，再脱蜡 5-10 min。脱蜡结束后，使用 100% 乙醇 5 min；接着用 90% 乙醇 2 min；最后用 70% 乙醇 2 min；DDH₂O 2 min。
2.2 冰冻切片：蒸馏水 2 min。
2.3 组培细胞：用4%多聚甲醛固定 10 min 以上。蒸馏水洗涤 2 min，换用新鲜的蒸馏水，再洗涤 2 min。
注：可根据具体实验选择其他的固定剂。
3.结晶紫染色
3.1 待检样本经以上方法处理后，直接加入结晶紫染色液室温染色 10 min (染色液需要覆盖住样本，具体的染色时间可根据需要的染色结果进行调整)。
3.2 倒去染色液，用蒸馏水充分洗涤直至没有染料被洗下来。
3.3 室温晾干。
3.4 光学显微镜下观察并拍照。
