Indocyanine green (solution) 

Indocyanine green (Foxgreen) (solution) is a low toxicic fluorescent agent that has been widely used in medical diagnostics, such as determining cardiac output, hepatic function, and liver blood flow, and for ophthalmic angiography (Ex/Em = 785/813 nm) ^[1].

1. 溶液配制^[3]
1.1 储备液保存
储存：分装后避光保存于 -20°C 或 -80°C。避免反复冻融。
1.2 工作液配制
用台氏液或其他缓冲液稀释储备液 (根据实验优化)。可根据实际情况稀释相应的储备液。注意，如果溶剂为 DMSO，则必须考虑 DMSO 的细胞毒性，并应制备溶剂对照；如果溶剂为纯水，则工作液在加入细胞前需要过滤并除菌。
注意：工作液应立即配制并使用，并避光保存。

2. 组织染色
2.1 实验采用新西兰白兔Langendorff灌注心脏。
2.2 将 Blebbistatin (20?μM) (HY-13813) 和 Biacetyl monoxime (5?mM) (HY-Y0413) 添加到灌注液中，以抑制心脏收缩，同时保持电活动持续。
2.3 将浓度逐渐增加 (从 0.03 μM 到 10 μM) 的 Indocyanine green 染料添加到灌注液中，对心脏进行染色。
2.4 用光谱仪记录的光信号持续时间为 3 秒。
