LysoTracker Red (solution) 

LysoTracker Red (solution) is a Red fluorescently labeled lysosomal probe with a maximum excitation/emission wavelength of 577/590 nm. The structure is composed of a fluorescein group and linked weak bases, which can freely cross the cell membrane and gather on spherical organelles. It is suitable for observing the internal biosynthesis and related pathogenesis of lysosomes^[1].

LysoTracker Red 工作液配制
1. 工作液配置：
1.1 将溶液恢复至室温，瞬时离心浓缩于管底。
1.2 用培养基或适当的缓冲液 (如PBS) 将1 mM 的储存液稀释到工作液中。推荐的工作溶液为 50-100 nM；
注意：请根据实际情况调整 LysoTracker Red 工作液的浓度，即配即用。
2. 细胞染色
2.1 悬浮细胞：4°C，1000g 离心，3-5 分钟，然后弃上清液。PBS 洗涤 2 次，每次 5 分钟。
贴壁细胞：弃细胞培养基，加入胰蛋白酶解离细胞，制成单细胞悬液。4°C 1000 g 离心 3-5 分钟，弃上清。PBS 洗 2 次，每次 5 分钟。
2.2 加入1 mL LysoTracker Red 工作液，室温孵育 5-30 分钟。
2.3 4°C，400g 离心 3-4 分钟，弃上清。
2.4 PBS 洗涤 2 次，每次 5 分钟。
2.5 用无血清细胞培养基或 PBS 重悬细胞，荧光显微镜或流式细胞仪检测。
