Sulfo-CY5-NHS ester potassium  (Synonyms: Sulfo-Cyanine5 NHS ester potassium)

Sulfo-CY5-NHS ester potassium is a reactive dye and can be used for labeling protein nucleic acid^[1].

Protocol
一、蛋白制备
1) 为获得最佳标记效果，请制备蛋白 (抗体) 浓度为 2 mg/mL。
2) 蛋白溶液的 PH 值为 8.5±0.5。如果 pH 值低于 8.0，应使用 1 M 碳酸氢钠进行调节。
3) 如果蛋白浓度低于 2 mg/mL，标记效率会大大降低。为获得最佳标记效率，建议终蛋白浓度范围为 2-10 mg/mL。
4) 蛋白必须在不含伯胺 (如 Tris 或甘氨酸) 的缓冲液中，并且铵离子，否则会影响标记效率。
2.染料制备 (以 Sulfo-Cyanine5 NHS ester 为例)
将无水 DMSO 加入 Sulfo-Cyanine5 NHS ester 小瓶中，制成 10 mM 储备液。通过移液器或涡旋混合均匀。
3.染料用量的计算
反应所需的 Sulfo-Cyanine5 NHS ester 的量取决于待标记蛋白的量，Sulfo-Cyanine5 NHS ester 染料与蛋白的最佳摩尔比为 10 左右。
Example：假设所需的 marker protein 为 500 μL 2 mg/mL IgG (MW=150,000)，用 100 μL DMSO 溶解 1 mg Sulfo-Cyanine5 NHS ester，得到所需的 Sulfo-Cyanine5 NHS ester 体积为 4.96 μL，详细计算过程如下：
1) mmol (IgG) =mg/mL (IgG) ×mL (IgG)/MW (IgG) =2 mg/mL × 0.5 mL/150,000 mg/mmol= 6.7×10^-6 mmol
2) mmol (Sulfo-Cyanine5 NHS ester) =mmol (IgG) × 10=6.7×10^-6 mmol×10=6.7 × 10^-5 mmol
3) μL (Sulfo-Cyanine5 NHS ester) = mmol (Sulfo-Cyanine5 NHS ester) ×MW (Sulfo-Cyanine5 NHS ester)/mg/μL (Sulfo-Cyanine5 NHS ester) =6.31 ×10^-5 mmol × 739.85 mg/mmol/0.01 mg/μL=4.96 μL (Sulfo-Cyanine5 NHS ester)
注意：我们建议使用 10:1 的染料/蛋白质摩尔比。如果浓度过低或过高，可将比例调整为 5:1、15:1 或 20:1。
4.运行偶联反应
1) 将适量新鲜制备的 10 mg/mL Sulfo-Cyanine5 NHS ester 缓慢加入到 0.5 mL 蛋白质样品中
在溶液中，轻轻摇动混匀，然后短暂离心，将样品收集在反应管底部。请勿混匀，以免蛋白样品变性失活。
2) 将反应管置于避光处，在室温下间隔轻轻孵育 60 分钟。10-15 分钟，轻轻逆转反应
5.纯化偶联物
以下方案是使用 SepHadex G-25 柱纯化染料-蛋白偶联物的示例。
1) 根据制造说明准备 SepHadex G-25 柱。
2) 将反应混合物加载到 SepHadex G-25 柱的顶部。
3) 一旦样品在顶部树脂表面下方运行，立即添加 PBS (pH 7.2-7.4)。
4) 向所需样品添加更多 PBS (pH 7.2-7.4) 以完成柱纯化。合并含有所需染料-蛋白质偶联物的级分。
