Methoxy-X04 (solution) 

Methoxy-X04 (solution) is a fluorescent dye that crosses the blood-brain barrier and selectively binds to beta-pleated sheets found in dense core amyloid Aβ plaques. Methoxy-X04 retains in vitro binding affinity for amyloid b (Ab) fibrils (K_i= 26.8 nM). Methoxy-X04 is fluorescent and stains plaques, tangles, and cerebrovascular amyloid in postmortem sections of AD brain with good specificity^[1][2].
Solvent and concentration: DMSO: 5 mg/mL

Methoxy-X04 (5-10 mg/kg，腹腔注射，一次) 可在 30 至 60 分钟内区分 PS1/APP 小鼠的单个斑块^[2]。

1. Methoxy-X04 溶液注射
灌注前 24 小时，称量小鼠体重，并给予每只小鼠腹膜内注射 Methoxy-X04，剂量为 10 mg/kg。
2. 注射小鼠的心脏灌注
3. 使用振动切片机进行脑切片
4. 切片筛查 Methoxy-X04 染色斑块的存在
为避免切片干燥，请按以下步骤依次检查每个切片：
用一次性移液器将一滴 PBS 滴到显微镜载玻片上，然后将切片放在 PBS 滴上。
在荧光显微镜 (激发波长340-380 nm) 下检查切片，以识别含有 Methoxy-X04 标记的 Aβ 斑块的区域。
