METTL3-IN-11 is an excellent, selective METTL3 inhibitor (IC50 = 45.31 nM). METTL3-IN-11 exhibits high selectivity towards METTL3 compared to DNMT1, EZH1, MLL1, and PRMT1. METTL3-IN-11 reduces the m6A level of total RNA in MOLM-13 and SKOV3 cells, induces cell apoptosis, and inhibits cell migration. METTL3-IN-11 can reduce the expression of m6A downstream target genes (c-MYC and BCL2). METTL3-IN-11 can be used for the study of ovarian cancer and acute myeloid leukemia[1].

Cell Migration Assay [1]

Incubation Time: 48 h
Result: Significantly inhibits the migratory capability of SKOV3 cells.

Western Blot Analysis[1]

Cell Line: SKOV3 cells, MOLM-13 cells
Concentration: 10 μM, 25 μM
Incubation Time: 48 h
Result: Did not affect the expression level of METTL3 protein.
Downregulated the protein expression of downstream target genes of m6A (such as c-MYC and BCL2).

METTL3-IN-11 (Compound 30t) (50 mg/kg,腹腔注射,每日一次,持续 2 周) 可显著抑制 SKOV3 荷瘤小鼠的肿瘤生长,并在实验期间保持体重稳定,且未观察到明显的毒性反应[1]

Animal Model: SKOV3 cells were subcutaneously inoculated into the right flank of female BALB/c mice (4–5 weeks old)[1].
Dosage: 50 mg/kg
Administration: I.p., once daily for 2 weeks
Result: Significantly inhibited tumor growth.
No significant difference was observed in the body weight.
