MitoPerOx (solution) is a mitochondrial-targeted, lipid peroxidation-indicating fluorescent probe with BODIPY581/591 fluorophores. The triphenylphosphine cation (TPP+) of MitoPerOx can be selectively enriched in mitochondria (depending on membrane potential) and can be used to detect lipid peroxidation in the inner mitochondrial membrane. Under the action of lipid peroxides, the BODIPY581/591 fluorophores of MitoPerOx shift their emission wavelength from 590 nm (reduced state) to 520 nm (oxidized state) , and ratiometric detection can be performed at an excitation wavelength of 488 nm. MitoPerOx can specifically monitor the peroxidation of mitochondrial phospholipids (especially cardiolipin) and is used in the study of oxidative stress-related diseases (such as aging, neurodegenerative diseases, and mitochondrial dysfunction) [1][2].

1.1 储备液
储存:分装后,置于 -20°C 或 -80°C 避光保存。避免反复冻融。
1.2 工作液配制
用 PBS 或细胞培养基稀释至 (根据实验优化)。可根据实际情况稀释相应的储备液。注意,如果溶剂为 DMSO,则必须考虑 DMSO 的细胞毒性,并应制备溶剂对照;如果溶剂为纯水,则工作液在加入细胞前需要过滤并除菌。
注意:工作液应立即配制并使用,并避光保存。


MitoPerOx 检测实验步骤细胞培养
将细胞 (如 HEK293 或成纤维细胞) 接种于 35 mm 培养皿的盖玻片上,于 37 °C、5% CO2 条件下培养。
探针准备:向无酚红培养基中加入 100 nM MitoPerOx,避光 37 °C 孵育 30 min。
洗涤:弃培养基,用 PBS 洗涤细胞 3 次。
诱导氧化应激:在 Hepes-Tris 缓冲液中加入 500 μM H2O2 处理细胞 15-30 min。
荧光成像:使用共聚焦显微镜 (激发波长 488 nm) 采集 520 nm (氧化态) 和 590 nm (还原态) 的发射光,通过计算 520/590 nm 比率量化脂质过氧化程度。
