Ovalbumin, low endotoxin, the main protein found in egg whites, have various biological activities such as anticancer, antihypertensive, antibacterial, antioxidant and immunomodulatory activities. Ovalbumins are the most abundant proteins synthesized in progesterone- or estrogen-treated fallopian tubes and are commonly used as markers to study hormone regulation of gene expression in tissues[1]. Ovalbumin, a low endotoxin, can be used to establish an asthma model[2][3].

致病原理
通过人工致敏和激发，在小鼠 (或其他动物，如大鼠) 体内模拟人类过敏性哮喘的 Th2 型免疫反应及其后续的炎症过程。
具体造模方法：
小鼠：BALB/c 小鼠 • 雌性 • 6-8 周龄
给药方式：100 μg • 腹腔注射 • 第 0、7 和 14 天，同时给予 2.25 mg Aluminum hydroxide (HY-B1521) • 然后于第 22、24 和 26 天吸入 Ovalbumin (OVA) (每只小鼠100 µg，溶于 50 µL PBS中)。
大鼠：SD 大鼠 • 雄性 • 4 周龄
给药方式：20 mg/kg • 腹腔注射 • 隔日一次，共注射 7 次 • 16 天后，大鼠每日接受 20 mg/kg OVA 超声雾化吸入，连续 7 天。雾化吸入 OVA 七天后，将 50 μL 2% OVA 滴入大鼠鼻腔，检测过敏性鼻炎 (AR) 反应。

Tips:
1) BALB/c 小鼠于最后一次 OVA 激发后 24 小时处死。
造模成功指标
行为分析：OVA 致敏导致大鼠频繁揉鼻子和打喷嚏。模型大鼠的 OVA 特异性 IgE 和 IgG1 水平较高。
气道高反应性：OVA 诱导的小鼠呼吸系统阻力 (Rrs) 和弹性 (Ers) 值显著升高，顺应性 (Crs) 值显著降低。
组织学分析：诱导支气管周围和血管周围区域浸润细胞总数显著增加。肺组织中以嗜酸性粒细胞为主。
OVA 致敏大鼠肺组织可见明显炎性细胞浸润和杯状细胞增生。
生化指标：OVA 致敏大鼠血清、NALF、BALF 及肺组织匀浆中 IL-17A 含量升高。
免疫组织化学分析：肺组织中 IL-22RA1 和 IL-10RB 表达显著增加。
