PTO-41 

PTO-41 is a blood-brain barrier-permeable near-infrared fluorescent probe that targets β-amyloid oligomers (Aβ Oligomers, AβOs) with a K_d of 349 nM. PTO-41 exhibits low cytotoxicity, high sensitivity to β-amyloid oligomers in in vitro phantom imaging, and can be rapidly cleared from the brain. PTO-41 is applicable to the research of Alzheimer's disease (Ex/Em = 538 nM/680 nM)^[1].

PTO-41 (25 μM) 可在与大脑相关的生物环境中选择性结合 Aβ42 寡聚体，产生的近红外荧光信号显著强于与 Aβ42 单体或聚集体结合时的信号^[1]。

PTO-41 (4 mg/kg；静脉注射；单次推注) 可穿透血脑屏障，并特异性标记 4 月龄 APP/PS1 小鼠体内的 β-淀粉样蛋白寡聚体；注射后 30 min 时，转基因小鼠的 NIRF 信号较野生型对照高 1.30 倍^[1]。

1. PTO-41 储备液配制
称取 PTO-41，溶于 DMSO 中。配制成 25.0 μM 储备液，避光，-20℃ 保存。
2. Aβ 物种荧光结合测试
2.1 用 PBS (pH 7.4) 将 PTO-41 稀释至终浓度 500 nM 于石英比色皿。
2.2 加入制备好的 Aβ42 单体/寡聚体/聚集体 (终浓度 1.5 μM，即探针:Aβ ≈ 1:3)。
2.3 混匀后室温孵育适量时间 (通常数分钟至平衡)。
2.4 测定荧光：Ex = 570 nm，Em scan = 600-800 nm (记录 680 nm 处强度)。
2.5 以不加 Aβ 的 PTO-41/PBS 为空白校正。
3. 小鼠活体 NIR 荧光成像
3.1 试剂配制 (注射液)：
溶剂体系：15% DMSO + 15% Cremophor EL + 70% PBS
PTO-41 终浓度按 4 mg/kg 体重计算 (通常 100 μL 用于小鼠)
临用前配制，摇匀静置 20 min 确保溶解稳定
3.2 动物成像
3.2.1 使用 4月龄雌性 APP/PS1 转基因鼠 及同龄 WT 对照鼠 (n=3-4)。
3.2.2 术前剃除头部毛发以减少背景。
3.2.3 尾静脉注射 100 μL PTO-41 溶液。
3.2.4 使用 IVIS Spectrum 系统采集：
Ex = 570 nm / Em = 680 nm
采集时间点：注射前、30、60、120、240、360 min
在软件 (Living Image 4.2.1) 中于脑部绘制 ROI 定量分析。
