生物活性

QX008N is a humanized IgG1 monoclonal antibody and also a TSLP inhibitor with a KD value of 0.0175 nM. QX008N blocks TSLP-induced signaling pathways by inhibiting the interaction of TSLP-TSLPR-IL-7Rα. QX008N reduces eosinophil and IgE levels, improves pulmonary function, blocks TSLP-induced inflammation, and enhances the inhibitory effect on inflammatory mediators when administered in combination with anti-IL-4R antibodies. QX008N is applicable to relevant research on asthma[1][2].

Animal Model: Cynomolgus monkey (male, 3-6 years, 2.5-7.0 kg, sensitized via intraperitoneal injection of Ascaris suis extract mixed with Al(OH)3 adjuvant at weeks 1, 2, and 3, followed by nebulized allergen challenge at weeks 4-7)[1]
Dosage: 10 mg/kg; 50 mg/kg
Administration: once weekly; 6 doses; subcutaneous injection
Result: Decreased percentage change in airway resistance by 25% and increased percentage change in lung dynamic compliance by 20% at one week after three 10 mg/kg doses.
Reduced peripheral blood specific IgE by ~38% after three 10 mg/kg doses and TSLP by ~42% after six 10 mg/kg doses.
Decreased percentage change in airway resistance by 42.4% and increased percentage change in lung dynamic compliance by 30% at one week after first 50 mg/kg dose.
Reduced peripheral blood TSLP by ~66% and specific IgE by ~62% after first 50 mg/kg dose.
Lowered TSLP levels, eosinophil counts and eosinophil percentage in bronchoalveolar lavage fluid at 10 mg/kg and 50 mg/kg.
Attenuated eosinophil infiltration in lung tissue at 10 mg/kg and 50 mg/kg.
Reached plasma concentration of 336.3 μg/mL after first 50 mg/kg dose.
