DEL技术通过将每个小分子化合物与一个独特的DNA序列共价连接，实现在一个管子里面同时筛选上百万或者上亿的化合物。传统的DEL筛选主要是发现非共价结合分子，这些分子与靶点之间的相互作用是可逆的。然而，共价DEL（CoDEL）则是在化合物中引入具有亲电反应活性的共价弹头，这些弹头能够与靶蛋白上特定的氨基酸残基（如半胱氨酸、赖氨酸等）形成稳定的共价键，进而增强结合强度，延长其在靶点上的停留时间，并有可能克服传统非共价抑制剂面临的耐药性或脱靶等效应。

本库是一个先进的、高度多样的化合物集合，由35 个独立子库组成，总分子规模达1400万，涵盖超过14个经过试验验证的共价弹头，这些弹头可靶向半胱氨酸，赖氨酸，精氨酸，天冬氨酸和谷氨酸。通过构建多样化核心成药骨架并整合可控共价反应弹头进行建库，兼具结构多样性、反应特异性和靶点广谱性等特点，可用于高效快速发现高选择性共价抑制剂。
