Sigvotatug vedotin (SGN-B6A; PF-08046047) is an ADC that targets integrin β-6 (ITGB6) via the human IgG1 monoclonal antibody Sigvotatug (HY-P990764) and exhibits cytotoxicity against a variety of integrin β-6-positive cancer cells through the mitotic inhibitor MMAE (HY-15162)[1].

(In Vitro)

Sigvotatug vedotin (2 μg/mL；4 h) 可与 BxPC-3 细胞的质膜结合，并在 37 °C 孵育 4 小时内通过受体介导的途径内吞进入溶酶体[1]。
Sigvotatug vedotin (96 h) 可在表达整合素 β-6 的 BxPC-3、Detroit 562、HPAF-II 和 SW780 癌细胞系中诱导抗原依赖性细胞毒性[1]。
Sigvotatug vedotin (0.001-50 μg/mL) 在 SW780 膀胱癌细胞中介导抗体依赖性细胞吞噬作用，其 EC50 为 0.7 μg/mL，但不会诱导抗体依赖性细胞毒性或补体依赖性细胞毒性[1]。

Result: Localized as a halo of red staining on the BxPC-3 cell plasma membrane at time 0.
Colocalized with green-stained lysosomes (appearing as orange color in intracellular vesicles) by 4 hours.
