生物活性

TSQ (solution) is a cytosolic zinc ion fluorescence probe that is membrane permeable and can be used for intracellular imaging of zinc proteins (λmax ~470 nm). TSQ can combine with Zn2+ in the presence of Ca2+ and Mg2+ to produce blue fluorescence (Ex/Em = 334/495 nm) [1].

1. 溶液配制[4]
1.1 储备液
储存：分装后避光保存于 -20°C 或 -80°C。避免反复冻融。
1.2 配制工作液
用 DPBS 或其他缓冲液稀释。可根据实际情况稀释相应的储备液。注意，如果溶剂为 DMSO，则必须考虑 DMSO 的细胞毒性，并应制备溶剂对照；如果溶剂为纯水，则工作液在加入细胞前需要过滤并除菌。
注意：工作液应立即配制并使用，并避光保存。

2. 细胞染色
2.1 将细胞培养于涂有 Collagen, rat tail (HY-NP010) 的玻璃盖玻片上。
2.2 用杜氏磷酸盐缓冲液冲洗三次。
2.3 将 30 μM TSQ 溶于 DPBS 中，充分混匀，并在 37°C 下孵育 30 分钟。
2.4 染色后的细胞用 DPBS 冲洗三次。
2.5 荧光显微镜检测 (激发/发射 = 334/495 nm)。
