WH-15 is a fluorogenic PLC reporter with Km value of 49; 30, 86.1 µM for PLC-γ1, PLC-δ1, PLC-β2, respectively. WH-15 can be cleaved in a cascade reaction to generate fluorescent 6-aminoquinoline. WH-15 can be used to respond to imaging PLC activity in live cells (Ex/Em = ~344/530 nm)[1].

(In Vitro)

1. 溶液配制[1]
1.1 原液配制
溶剂：DMSO
浓度：25 mM (根据实验优化)。
储存：分装后避光保存于 -20°C 或 -80°C。避免反复冻融。
1.2 工作液配制
用检测缓冲液稀释至 50 µM。
注意：工作液应立即配制并使用，并避光保存。

2. 细胞染色
2.1 HEK-293T 细胞转染 600 ng DNA 和 100 ng 空载体。
2.2 将 70 μL 50 μM WH-15 (83 μg/mL 无脂肪酸 BSA、50 mM HEPES (pH 7.2)、70 mM KCl、3 mM CaCl2、3 mM EGTA 和 2 mM DTT) 加入 50 μL 标准化细胞裂解液中，37°C。
2.3 使用多标记计数器 (Ex/Em = 340/535 nm) 每 3 分钟采集一次数据。
