X-GAL (solution) 

X-GAL (BCIG) (solution) is a widely used chromogenic β-galactosidase substrate. X-GAL is a colorless compound until cleaved by β-galactosidase, at which point X-GAL turns to an insoluble and detectable blue compound, making X-GAL particularly useful in techniques such as blue-white screening for cloning in bacteria. X-GAL can also be used for detection of β-galactosidase activity^[1][2].
Solvent and Concentration: DMF: 20 mg/mL

1. 溶液配置^[1]
1.1 储存液
保存：分装后 -20°C 或 -80°C 避光保存，避免反复冻融。
1.2 工作液的配制
用 1 mL 琼脂培养基将 X-gal 稀释至 40 μg/mL，并添加 1 μL 的 IPTG (HY-15921) (24 mg/mL) 和 1 μL 的 AMP (HY-A0181) (100 mg/mL) (根据实验优化)。
注意：工作液现配现用，避光。

2. 蓝/白菌落筛选
2.1 当 DNA 片段插入至 pUC 系列载体 (或其他带有 lacZ、Amp 基因载体) ，然后转化至 lacZ 缺失细胞中后，涂布上述的 X-gal、IPTG、AMP 平板培养基。
2.2 37°C，直至琼脂表面出现蓝白色菌落。
2.3 将蓝白色菌落进一步培养，并通过双酶切鉴定。
2.4 吸去染料工作液，用 PBS 洗 2-3 次，使用光学显微镜进行观察。

将细胞 (以 HUVEC 为例) 在培养皿中培养至 80% 的合度时，在培养基中加入 50 μmol/L H₂O₂，培养 12 h^[2]。
随后用胰蛋白酶/EDTA 收获细胞，用 PBS 洗涤 3 次，再用 4% 甲醛固定 10 min。
以同样方法制备未经 H₂O₂ 处理的细胞作为对照。
37 °C 下 X-gal 染色。

切除肠组织，纵向切开。将肠道内容物用磷酸盐缓冲液冲洗干净后，将肠道组织内表面朝上置于载玻片上^[2]。
将含有 β-gal (大肠杆菌 DH5α 含 pUC18) 的单个蓝色菌落在 100 μL PBS 中稀释，然后在组织中加入 10 μL。
孵育 1 小时后，用 PBS 溶液洗涤组织。作为对照，用同样的方法制备不含蓝色菌落的组织。
所有组织用 4% 甲醛固定 10 分钟，并在 37 °C 下加入 X-gal 进一步染色。
