Abstract

Abstract A general method of overcoming the original specificity of proteases is illustrated by a combination of the substrate mimetic strategy shown here with the broad S′-subsite specificity of the protease clostripain. Besides the synthesis of peptide isosteres and analogues, the biocatalytic approach presented allows the coupling of completely non- amino-acid-derived carboxy and amino components.
