General procedure for the synthesis of 7-chloro-6-aminobenzothiazole from 6-aminobenzothiazole: To a 100 mL solution of 6-aminobenzothiazole (2.0 g, 13.4 mmol) in acetic acid (AcOH) was added a solution of acetic acid saturated with chlorine (Cl2). The reaction mixture was stirred at 25°C for 1 hour. After completion of the reaction, the mixture was concentrated in vacuum to give a yellow solid. The solid was dissolved in ethyl acetate (EtOAc) and washed with aqueous sodium bicarbonate (NaHCO3). The organic layer was dried with anhydrous sodium sulfate (Na2SO4) and concentrated under vacuum to give an oil. Finally, purification by column chromatography (eluent: 10-50% ethyl acetate/hexane) gave 7.8 g (34 mmol, 80% yield) of 7-chloro-6-aminobenzothiazole.
