Western blot analysis of extracts from C6(lane 2(20ug) , NIH/3T3(lane 3(20ug) and RAW264.7(lane 4(20ug) using Annexin-2/ANXA2 Antibody (HY-P80015) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
Immunocytochemistry analysis of Hela cells labeling Annexin-2/ANXA2 with Annexin-2/ANXA2 Antibody (HY-P80015) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with Annexin-2/ANXA2 Antibody (HY-P80015) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

背景

功能：钙调节膜结合蛋白，其对钙的亲和力可被阴离子磷脂显著增强。它能以高亲和力结合两个钙离子。可能参与热应激反应。抑制 PCSK9 增强的 LDLR 降解，可能通过翻译机制降低 PCSK9 蛋白水平，但也会与 LDLR 竞争结合 PCSK9 (PubMed:18799458, PubMed:22848640, PubMed:24808179)。结合被颗粒磨损碎片吞噬作用损伤的内体，参与内体膜稳定，从而限制 NLRP3 炎症小体的激活 (基于相似性)。内皮细胞表面纤溶酶生成所必需，可能支持纤溶监视和新生血管生成 (基于相似性)；(微生物感染) 结合肺炎支原体 CARDS 毒素，可能作为该病原体的受体之一。当 ANXA2 被 siRNA 下调时，毒素与人体细胞的结合减少，空泡化 (肺炎支原体感染的症状之一) 也随之减少。

亚基：异源四聚体，包含 2 条 S100A10/p11 轻链和 2 条 ANXA2/p36 重链 (通过相似性)。
