CD4 Antibody (YA537) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to CD4.

CD4 蛋白在细胞表面形成二硫键连接的同二聚体，并与多种蛋白相互作用，包括 LCK、PTK2/FAK1 和 P4HB/PDI。它还与 IL16 结合，导致淋巴细胞中 CD4 依赖性信号传导。此外，CD4 与 MHCII α 和 β 链相互作用，增强 T 细胞受体 (TCR) 对肽-MHCII 的亲和力。CD4 的寡聚化，特别是通过 Ig 样 C2 型结构域 2 和 3 进行的寡聚化，对于稳定结合 MHCII 并促进 T 细胞和抗原呈递细胞 (APC) 之间的粘附至关重要。

  Western blot analysis of extracts from THP-1(lane 2(20μg), U937 (lane 3(20μg) and HEK293(lane 4(20μg) using CD4 (HY-P80055) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

  Immunocytochemistry analysis of Hela cells labeling CD4 with CD4 Antibody (HY-P80055)at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with CD4 Antibody (HY-P80055) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).
