Transposable elements are regarded as a powerfulmutagen and as an effective tool to isolate genes taggedby transposon insertions. The Japanese morning glorycontains around 500-1000 copies of an En/Spm-relatedelement Tpn1 and its relatives, which act as majorspontaneous mutagens. We have previously developedan amplified restriction fragment length polymorphism(AFLP)-based mRNA fingerprinting (AMF) procedurewhich is based on the systematic comparison of differentlyexpressed transcripts in the same tissue in differentlines, and succeeded in applying AMF for the identificationof a new mutable allele caused by integrationof a transposable element into an anthocyanin biosynthesisgene. Since transposon mutagenesis has become apowerful tool for the isolation of genes of interest, wehave attempted to develop a new protocol for identifyingtagged genes by insertion of Tpn1-related elementsin the Japanese morning glory. Our transposon taggingmethod, named simplified transposon display (STD),was based on our AMF procedure and is simple andrequires neither biotinylated oligonucleotides norstreptavidin-capturing which are essential in othertransposon display methods published recently.
