人类肝脏细胞（HHpC）分离自单个正常供者的肝脏，3 - 6×106 冻存细胞，货号C-12850。分离后，细胞立即冻存在液氮中，然后进行全面的质量检验。检验的项目有细胞活性、细胞附着率、I期代谢、II期代谢和白蛋白合成。细胞附着率至少为50%。
肝脏是机体中的主要代谢器官，执行糖酵解、糖原生成、氨基酸代谢、脂质代谢和尿素生成的功能。此外，肝脏也参与调控血糖水平。同时，由于来自胃肠道的富含营养的血液首先要通过肝脏，肝脏细胞能在吸收毒性物质之后很快将其清除。这整个解毒过程作用包括I期反应和II期两步反应。I期反应的酶系统中最重要的是细胞色素P450系统。因此，体外培养的肝脏细胞可以完美地适用于临床前或临床研究之前的体外代谢和毒性/解毒过程研究。
注意：在标准的体外细胞培养条件下，成熟的人类肝脏细胞通常存活不会超过10-14天，且不会增殖。 The cells have been tested: Albumin positive.

How can I culture the human hepatocytes? Do they adhere to TC plastic?
Our human hepatocytes (HHpC) can either be maintained in suspension culture for a few hours or cultured in collagen type I-coated tissue culture vessels for about 2-3 weeks.
We guarantee > 50% attachment rate when PromoCell Hepatocyte Growth Medium (C-25010) is used. The recommended seeding density after thawing is 150,000 viable cells/cm² and the first media change should be performed after 4 hrs.
What is the difference between Hepatocyte Growth Medium (C-25010) and Hepatocyte Maintenance Medium (C-25020)?
The PromoCell Hepatocyte Growth Medium is a serum-free medium that contains all the growth factors and supplements necessary for longterm cultivation of hepatocytes. It is used to plate the cells after thawing.
In case some of the supplements interfere with the experimental setups, you can switch to Hepatocyte Maintenance Medium after attachment. This medium is supplemented with insulin and dexamethasone. The culture time in the Maintenance Medium should not exeed 3 days.
Do the human hepatocytes proliferate in vitro?
No, human hepatocytes do not considerably proliferate in culture. They remain attached for approx. 2 weeks when cultured in our Hepatocyte Growth Medium (C-25010). 
As most scientists are interested in "fresh" hepatocytes with high metabolic capacity, the tests are usually performed shortly after recovery and finished within a few hours to days.
Where is the tissue for hepatocyte isolation sourced from?
The liver tissue is sourced from visceral surgery. It comes from patients undergoing surgery due to a liver tumor. During the operation, the tumor is removed together with some surrounding non-tumor tissue. This non-tumor tissue is used for hepatocyte isolation and the cells are cryopreserved immediately after isolation.
Is it necessary to use coated flasks for the culture of primary human hepatocytes?
Yes, we recommend to use collagen type I coated flasks.
Short protocol for surface coating:
Dilute the needed amount of Type I Rat Tail Collagen Solution to 50 µg/ml with 0.01M HCl. For surface coating of tissue culture plasticware add 5-10 µg/cm² diluted collagen solution to the culture surface. Be sure that the entire surface is covered. Place flasks on a level surface at room temperature for 60 min. Aspirate the collagen solution. Rinse well with at least 4 volumes of sterile PBS or medium. Aspirate the PBS. Use immediately or leave flasks uncapped in the hood to dry. 
Unused vessels should be capped and can be stored at 4°C for up to 2 weeks.
