背景资料	Myeloperoxidase (MPO) is a mammalian phagocyte hemoprotein thought to primarily mediate host defense reactions. It is abundantly expressed in neutrophils and secreted during their activation. Myeloperoxidase is part of the host defense system of human polymorphonuclear leukocytes, responsible for microbicidal activity against a wide range of organisms. It is located in the nucleus as well as in the cytoplasm. Intranuclear MPO may help to protect DNA against damage resulting from oxygen radicals produced during myeloid cell maturation and function. The standard product used in this kit is the product of gene recombination, consisting of 697(A49-S745) amino acids with the molecular mass of 80KDa.

Lane 1: Recombinant Human Myeloperoxidase Protein 0.5ngAfter Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Myeloperoxidase antigen affinity purified polyclonal antibody (Catalog # PB9057) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Myeloperoxidase at approximately 37KD.
