Biglycan, also known as PG I is an extracellular matrix glycoprotein belonging to the small leucine-rich proteoglycan (SLRP) family. Biglycan consists of a protein core containing leucine-rich repeat regions and two glycosaminoglycan (GAG) chains consisting of either chondroitin sulfate (CS) or dermatan sulfate (DS). The composition of the GAG chains has been reported as varying according to tissue of origin. Biglycan is found in several connective tissues, especially in articular cartilages. Biglycan binds several matrix proteins, including fibrillar collagens, Matrilin1, and βIGH3. It also binds and modulates the activity of a variety of nonmatrix proteins, including BMP4, C1q, collectins, TGFβ, TNFα, and WISP1. Biglycan functions in the development and maintenance of many tissues. Studies suggest that biglycan plays important roles in osteoarthritis.It has been reported that recombinant extracellular domain of Biglycan decreases muscle pathology and improves muscle function in dystrophic mice.
·Description
Recombinant Human Biglycan produced by transiently transfected human cell is the secreted mature form(Accession#P21810) (Asp38-Lys368) of human biglycan fused with a polyhistidine tag at the C-terminus.
·Quality Control
Biological activity: As measured by its ability to increase the binding of BMP4 to Chordin, 2-8μg/mL of rhBiglycan has a 50% maximum increase in the binding of rhBMP4 (0.1 μg/mL) to rmChordin (1 μg/mL).
Purity: Greater than 90.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Amino-Acid Sequence: Predicted N terminal amino acid is Asp38.
Endotoxin:Less than 0.1ng/μg (1EU/μg) of rHu rHuBiglycan.
·Formulation
Human FGF-basic was lyophilized from 1mg/ml solution after extensive dialysis against 20 mM phosphate buffer, pH 7.2, 50 mM Na2SO4, 0.2 mM DTT and 0.2 mM EDTA.
·Reconstitution
It is recommended to reconstitute the lyophilized rHuBiglycan in sterile water not less than 100μg/ml, which can then be further diluted to other aqueous solutions.
·Storage
Lyophilized rHuBiglycan although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution rHuBiglycan should be stored at 4°C between 2-7 days and aliquots of reconstituted samples containing 0.1% BSA are stable up to 3 months below -18°C for future use.
