荧光标记法操作步骤:
1.用碳酸氢钠缓冲液pH9.8稀释抗体为1-5mg/m1,或抗体对该缓冲液充分透析,以足以使赖氨酸不解离(去其正电荷),但注意保持大部分蛋白质仍未变性;
2.将透析袋放入100m1含0.1mg/ MI fitc的pH9.8碳酸氢钠缓冲液(新配制)的烧杯中,用铝铂包烧杯以避光,4℃搅拌过夜;
3.上述抗体液对PBS在4℃透析以终止反应。其间至少更换PBS液三次,直致480nm的吸收为零;
4.加入0.5g/L的叠氮-钠。此后,结合物在4℃避光保存,或分装后在-20℃冻存。
荧光标记抗体实验要点:
1.偶联反应要求在尽可能接近pH9.8的溶液中进行,而且注意在反应过程中要保持此pH水平;
2.要确定在偶联反应缓冲液中不含游离氨基(Tris,氨及叠氮-钠均可与FITC反应,因此会降低蛋白质与FITC的偶联率);
3.FITC与蛋白质的比值(F/P),可通过测定495m与280nm吸光度来鉴定;
4.FITC缺限是易被光淬灭,因此复合物必须始终避光保存;
5.如果FITC一复合物对PBS透析不充分,可能造成高本底,对免疫荧光染色产生干扰;荧光标记法操作步骤:
