MLPA

MLPA在研究和诊断中的作用:
MLPA® (Multiplex Ligation-dependent Probe Amplification) is the go-to technique for studying gene copy number variations (CNVs) associated with disease. Laboratories worldwide rely on MLPA for the diagnosis and research of genetic disorders and tumours. The power of the technique lies in its versatility: MLPA can be used to detect copy number changes in anything from complete chromosomes to single exons. MLPA is also used to detect DNA methylation changes (MS-MLPA) and is sensitive enough to discriminate aberrations in disease-causing genes from highly similar pseudogenes.
选择MLPA的理由:
CNVs play a role in hundreds of genetic disorders, and MLPA assays exist for even the rarest of them. Laboratories around the world use MLPA to ensure that deletions and duplications are adequately recognised. For disorders where CNVs make up the majority of mutations, MLPA is used as a first-line test. The same applies to disorders in which genetic analysis is complicated by pseudogenes: here, MLPA is used for its exceptional sensitivity to discriminate closely related sequences. In oncogenomics, MLPA is used to study somatic CNVs to optimise tumour profiling.
MLPA的原理:
MLPA probesMLPA probemixes have probes that target a specific genomic sequence. An MLPA probe consists of two parts: a left and a right probe oligonucleotide (LPO and RPO). The LPO and RPO contain PCR primer and DNA hybridisation sequences. An additional stuffer sequence in the RPO gives each probe a unique length.
Sample denaturation and probe hybridisationIn the first step, purified sample DNA is denatured. This is followed by overnight incubation with MLPA probe oligos. The LPO and RPO parts of each probe hybridise to immediately adjacent target DNA sequences.
Probe ligationThe second step is the ligation of probe oligos that hybridised to immediately adjacent target sequences. No mismatches around the ligation site are permitted, making the ligation reaction highly specific. The number of probe ligation products is a measure for the number of target sequences in the sample.
Probe amplificationIn the third step, ligated probes are amplified in a multiplex PCR using a single universal primer pair. Only ligated probes are exponentially amplified, making the removal of unbound and non-ligated probes unnecessary.
Fragment separationThe fourth step is fragment separation by capillary electrophoresis. PCR products are loaded onto a capillary electrophoresis device and separated by length. Each fragment corresponds to a specific MLPA probe.
Data analysisThe final step is data analysis by Coffalyser.Net. Relative copy numbers are determined by comparing the relative peak heights of reference probes and target probes in the test samples with those in reference samples with a known normal copy number. Advanced quality checks help to recognize unreliable data.

digitalMLPA简介:
SALSA® digitalMLPA™ is similar to conventional MLPA, the gold standard for CNV detection of multiple DNA sequences, but with the ability to examine many more targets. digitalMLPA takes the guesswork out of NGS CNV calls by combining the MLPA technology you trust for CNV detection with the large number of targets you have come to expect with NGS. With digitalMLPA, you can examine more targets than with conventional MLPA, with decreased costs and turnaround times.

The first SALSA digitalMLPA panel, D001 Hereditary Cancer Panel 1, offers gene coverage that exceeds that of ten conventional MLPA assays, thereby streamlining oncogenetic screening.
digitalMLPA工作原理:
digitalMLPA probes
digitalMLPA probemixes have probes that target a specific genomic sequence. A digitalMLPA probe consists of two parts: a left and a right probe oligonucleotide (LPO and RPO). The LPO contains a P7 PCR primer and a DNA hybridisation sequence. The RPO contains a sequence for distinct digitalMLPA read identification and a DNA hybridisation sequence. A Barcode Oligo (BO) is added to every reaction. The BO contains a unique sequence for sample identification, along with Rd1 and P5 primer sequences.
Sample denaturation and probe hybridisation
In the first step, purified sample DNA is denatured. This is followed by overnight incubation with digitalMLPA probe oligos. The LPO and RPO parts of each probe hybridise to immediately adjacent target DNA sequences.
Probe ligation
The next step is the ligation of probe oligos that hybridised to immediately adjacent target sequences. No mismatches around the ligation site are permitted, making the ligation reaction highly specific. The number of probe ligation products is a measure for the number of target sequences in the sample. With digitalMLPA, it is possible to detect the number of ligation events.
Probe amplification
Ligated probes are amplified in a multiplex PCR using a single P7/P5 primer pair. Only ligated probes are exponentially amplified, making the removal of unbound and non-ligated probes unnecessary.
Amplicon sequencing
PCR products are mixed in equal amounts, diluted, denatured, and loaded onto an Illumina sequencer. For each probe in all reactions, at least 400 single reads should be generated for reliable quantification.
Data analysis
The final step is data analysis by Coffalyser digitalMLPA. Coffalyser digitalMLPA generates two easy-to-interpret reports for each sample in your experiment: one with general details and one with sample results. These reports indicate if sample and reaction quality were sufficient and highlight detected aberrant regions.
