pUCm-T载体是为简化PCR产物的克隆而设计的。许多热稳定的DNA聚合酶,如Taq DNA聚合酶、Tth DNA聚合酶等扩增的PCR产物在3’末端后都带有一个突出的碱基A,这样的PCR产物很容易连接到带有突出碱基T的T载体上。
pUCm-T载体是一种新颖的pUC系列T载体,其多克隆位点大多是单切点,以及β-半乳糖苷酶读码框的调整,大大方便了克隆的蓝/白筛选。插入位点两端独特设计的两个PstI位点使插入片段可以用Pst I单酶切进行检测,也可以用非常廉价而高效的EcoR I和Hind III双酶切进行检测。可以用M13通用引物和T7启动子引物对PCR产物进行测序。pUCm-T含有T7 RNA聚合酶的启动子,可以对插入片段进行体外转录。
