测定意义:土壤脱氢酶(Soil dehydrogenase, sDHA)的活性可以反映土壤体系内活性微生物量以及其对有机物的降解活性,可以作为土壤微生物的降解性能指标。
测定原理:氢受体2, 3, 5 - 氯化三苯基四氮唑(2,3,5-Triphenyl Tetrazolium Chloride, TTC)在细胞呼吸过程中接受氢以后,被还原为三苯基甲鐟(Triphenyl Formazone, TF),TF呈现红色,在波长485nm处有最大吸收峰,采用分光光度法于485nm测定其吸光值,即得土壤脱氢酶活性。
需要的仪器,耗材:筛子、天平、恒温培养箱或水浴锅、低温离心机、漏斗,滤纸、可见分光光度计、玻璃比色可见分光光度计/酶标仪、微量石英比色皿/96孔板、冰、蒸馏水、丙酮(不允许快递,请用户自备)。
