A series of N-phenyl thalidomide analogues have been prepared efficiently through a Buchwald–Hartwig cross coupling reaction as the key step. Several of these compounds have extraordinary TNF expression inhibition compared to both thalidomide and the current pharmaceutical agent Revlimid (Lenalidomide).
