ABBV-011 (SC-011) is a SEZ6-targeted, antibody-drug conjugate (ADC). ABBV-011 binds cell surface-expressed SEZ6 by anti-SEZ6 antibody Turmetabart (HY-P991041), triggers ADC-receptor complex internalization into lysosomes, releases Calicheamicin (HY-19609) payload, and mediates cytotoxicity. ABBV-011 induces tumor regression and mediates selective killing of SEZ6-positive cells. ABBV-011 can be used for the research of small cell lung cancer[1]. (Pink: Target protein ligand; Blue: E3 ligase ligand; Black: linker).

ABBV-011 (0.01-100000 pM; 96 h) 可强效且选择性地杀伤过表达人源 SEZ6 的 HEK293T 细胞，孵育 96 h 后其 IC50 为 75 pmol/L[1]。
ABBV-011 (1-100000 pM; 96 h) 可在亲本 NCI-H69 小细胞肺癌 (SCLC) 细胞中介导靶标特异性细胞毒性，孵育 96 小时后的 IC50 为 106 pmol/L，但在 SEZ6 敲除的 NCI-H69 细胞中无活性[1]。

Incubation Time: 96 h
Result: Had strong activity in parental NCI-H69 cells with an IC50 of 106 pM.
Showed no activity at the highest concentration tested in SEZ6-knockout NCI-H69 cells.

ABBV-011 (0.5-8 mg/kg；腹腔注射；单次给药) 可在 SEZ6 阳性小细胞肺癌 PDX 模型中诱导呈剂量依赖性的、靶向 SEZ6 的肿瘤消退[1]。

Animal Model: NOD/SCID with SCLC P PDX tumor cells (female, 5 to 10 weeks of age)[1]
Dosage: 0.5 mg/kg; 1 mg/kg; 2 mg/kg; 4 mg/kg; 8 mg/kg
Administration: i.p.; single dose
Result: Induced sustained tumor regression in SEZ6-positive LU64, LU95, LU129 and LU149 PDX.
Exhibited minimal effect on tumor growth in SEZ6-negative LU505 PDX even at 8 mg/kg.
