Fluorescein-5-maleimide (solution)  (Synonyms: N-(5-Fluoresceinyl)maleimide (solution))

Fluorescein-5-maleimide (solution) (N-(5-Fluoresceinyl)maleimide (solution)) is a fluorescent dye. Fluorescein-5-maleimide can be used to detect the redox state of thiols in eukaryotic cells. Fluorescein-5-maleimide can label peptides and is used to detect negatively charged nanoparticles. Fluorescein-5-maleimide can also label actin to explore its interaction with cardiac myosin-binding protein C (cMyBP-C), which helps in developing small molecule modulators for heart failure. Fluorescein-5-maleimide can screen mutant proteins that contain cysteine residues. The excitation wavelength of Fluorescein-5-maleimide is 494 nm, and the emission wavelength is 519 nm^[1][2][3][4].

Fluorescein-5-maleimide 用于检测细胞内硫醇氧化还原状态

Fluorescein-5-maleimide 荧光探针的配制^[2]
1.1 制备工作液

2) 将 1) 中混合物 (终体积为 125 μL) 在室温下黑暗孵育 2 小时，并轻轻摇动。
3) 加入过量的 DDT (20 mM) 来淬灭反应。

5) 将 5 μL 荧光探针添加到 15 μL 的纳米粒子和 130 μL 的缓冲溶液中，检测两者相互作用。

Fluorescein-5-maleimide 用于检测蛋白相互作用 (肌动蛋白与心肌肌球蛋白结合蛋白C (cMyBP-C))^[3]
1) 将 50 μM 的 G-肌动蛋白在 20 mM Tris pH 7.5、0.2 mM CaCl₂、0.2 mM ATP 的条件下，加入 3 M KCl (最终浓度为 100 mM) 和 0.5 M MgCl₂ (最终浓度为 2 mM) 进行聚合，在 23 °C 下孵育 1 小时。
2) 加入 Fluorescein-5-maleimide (终浓度为 1 mM) ，在 23 °C 下标记 1 小时。
3) 加入五倍摩尔、过量的 DTT 来停止标记。
4) 将 0.25 μM 的 Fluorescein-5-maleimide-肌动蛋白和 0-20 μM 的 TMR-cC0-C2 (cMyBP-C 的 N 末端片段) 共同孵育。
5) 数据分析 (FLT-FRET 测定)：首先激发 Fluorescein-5-maleimide 并检测其 FLT，以测量从 Fluorescein-5-maleimide (供体) 到 TMR (受体) 的荧光共振能量转移 (FRET) 的变化，Fluorescein-5-maleimide-肌动蛋白的 FLT 降低 (FRET 增加) 表明结合。
