生物活性

Green DND-26 (solution) is a green fluorescently labeled lysosomal probe with a maximum excitation/emission wavelength of 504/511 nm. The structure is composed of a fluorescein group and linked weak bases, which can freely cross the cell membrane and generally gather on spherical organelles. Green DND-26 is suitable for observing the internal biosynthesis and related pathogenesis of lysosomes[1].

Green DND-26 工作液的配制
一、储备液的配制
1.1 将溶液恢复至室温，瞬时离心浓缩至管底。
1.2 稀释使用介质或适当的缓冲液 (如 PBS) 将 2 mM 存储溶液添加到工作溶液中。推荐工作液浓度为 50-100 nM。
注意：请根据实际情况调整 Green DND-26 工作液的浓度，即用即用。
细胞染色
2.1 For 悬浮细胞：4℃，1000 g 离心 3-5 分钟，弃上清。PBS 洗涤2次，每次 5 分钟。
贴壁细胞：弃去细胞培养基，加入胰蛋白酶解离细胞，制成单细胞悬液。4℃ 1000 g 离心 3-5 分钟，弃上清。PBS 洗涤 2 次，每次 5 分钟。
2.2 加入1mL Green DND-26 工作液，室温孵育 30 分钟。
2.3 4℃ 400 g 离心 3-4 分钟，弃上清。
2.4 PBS 洗涤 2 次，每次 5 分钟。
2.5 用无血清细胞培养基或 PBS 重悬细胞，荧光显微镜或流式细胞仪检测流式细胞仪。
