Herpetin is an active lignan. Herpetin acts as a bone marrow mesenchymal stem cell inducer that activates the SDF-1/CXCR4 axis and the Wnt/β-catenin pathway. Herpetin is applicable to research related to acute liver injury[1][2].

Cell Viability Assay[1]

Cell Line: passage 3-5 mouse bone marrow mesenchymal stem cells (BMSCs)
Concentration: 10, 25, 50, 75, 100 μM
Incubation Time: 24, 48, 72 h
Result: Significantly reduced BMSC viability at 25, 50, 75, 100 μM after 24, 48, and 72 h.
Showed no significant effect on BMSC viability at 10 μM across all tested time points.

Cell Differentiation Assay[1]

Cell Line: passage 3-5 mouse bone marrow mesenchymal stem cells (BMSCs)
Concentration: 10 μM
Incubation Time: 7, 14, 21 d
Result: Significantly increased relative CYP3A4 fluorescence intensity compared to untreated BMSCs at 7, 14, and 21 d.
Significantly increased glycogen storage in BMSCs compared to untreated BMSCs at 14 and 21 d; no significant difference was observed at 7 d.
Increased alpha-fetoprotein (AFP), albumin (ALB), and cytokeratin 18 (CK18) expression compared to untreated BMSCs, with statistically significant increases at 14 and 21 d and non-significant increases at 7 d.
Peaked AFP expression at 14 d then decreased, while ALB and CK18 expression increased continuously from 7 to 21 d.

Administration: i.p.; single dose (administered 8 hours post-liver injury induction)
Result: Reduced serum AST levels compared to the model group.
Reduced the liver histopathological severity score compared to the model group.
Elevated liver tissue expression of SDF-1, CXCR4, p-GSK-3β, and β-catenin compared to the model group.
Improved mouse survival rate compared to the model group.
