M104 peptide is an OXA-48 carbapenemase inhibitor and antibiotic potentiator. M104 peptide effectively blocks the binding and active site cavity of OXA-48. M104 peptide restores Meropenem (HY-13678)’s antibacterial activity against OXA-48-producing Klebsiella pneumoniae. M104 peptide can be used for the research of Carbapenem-resistant Klebsiella pneumoniae infection[1][2].

M104 peptide (7-125 μg/mL; 18 h) 可使对 Meropenem 耐受、携带 blaOXA-48 的肺炎克雷伯菌菌株的 Meropenem MIC 降低 4 倍，恢复其对 Meropenem 的中介敏感性[1]。
Peptide M104 (50-200 μg/mL; 18 h) 可增强美罗培南对强生物膜形成能力的 blaOXA-48 阳性肺炎克雷伯菌临床分离株的生物膜抑制活性，使所有 5 株受试菌株的美罗培南 MBIC 值降低 2 倍，但整体效果无统计学显著性 (p-value = 0.06)[2]。
Peptide M104 (200 μg/mL; 24-48 h) 在孵育 24 h 后对 HDFa 细胞无细胞毒性，但在孵育 48 h 后会导致细胞活力显著降低 9.9%[2]。
Peptide M104 (200 μg/mL; 1 h) 对新鲜人红细胞孵育 1 小时后无溶血活性[2]。

Cell Cytotoxicity Assay[2]

Cell Line: Human dermal fibroblast (HDFa) cell line
Concentration: 200 μg/mL
Incubation Time: 24 h; 48 h
Result: Maintained a mean cell viability of 99.6% in HDFa cells after 24 h of incubation, which was not statistically different from the untreated control (p-value > 0.05).
Reduced mean cell viability significantly to 90.1% after 48 h of incubation compared to the untreated control (p-value < 0.05).
