MeDZ lipid is a zwitterion-type ionizable endosomal membrane destabilizer and anti-inflammatory agent that promotes endosomal escape. When incorporated into LNP formulations, MeDZ lipid enhances mRNA expression in lymph node antigen-presenting cells and promotes cytotoxic T cell activation. MeDZ lipid is compatible with existing targeted nanoparticle formulations to improve mRNA delivery efficiency[1].

MeDZ lipid (孵育 10 min；pH 7.4-5.5) 具有 pH 响应性质子化特性，在 pH ≥6.8 时为结合 H2O 的两性离子状态，在 pH <6.8 时转变为结合 H3O+的带正电状态，这一特性已通过 1H NMR spectroscopy 检测到[1]。

含 25 mol% MeDZ lipid (pH 6.0) 的 LNP 与对照 LNP 相比，在酸性 pH (6.0) 下表现出更强的膜破坏活性，这表明其具有更优异的内体逃逸能力[1]。
含有 25 mol% MeDZ lipid 的 LNP (15 μg/mL; 1.5 h incubation) 在 DC2.4 细胞中表现出更强的内体逃逸能力；与对照 LNP (PCC = 0.90) 相比，其与内体/溶酶体的共定位水平降低 (PCC = 0.78)[1]。
含有 25 mol% MeDZ lipid 的 LNPs (1 μg/mL OVA mRNA；6 h incubation, 24 h expression/presentation incubation) 可增强 BMDCs 中 OVA 抗原的表达与呈递，处理后 OVA 蛋白水平约为对照 LNPs 的 100%，且 H2Kb-SIINFEKL 阳性细胞占比为 16.0%[1]。
