MK-4721  (Synonyms: AGS-1C4D4; AGS-PSCA)

AGS-1C4D4 is a fully human IgG1κ monoclonal antibody targeting PSCA, with a K_a value of 0.2 nM. AGS-1C4D4 mediates antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) against PSCA-expressing cells. AGS-1C4D4 can be used for the research of pancreatic cancer^[1].

AGS-1C4D4 对表达 PSCA 的 HPAC 和 Panc0203 胰腺细胞系表现出强效的 ADCC 活性，去糖基化会使其活性大幅降低，且在该实验中其效力高于杂交瘤来源的 AGS-PSCA 抗体^[1]。
AGS-1C4D4 对表达 PSCA 的重组 B300.19 细胞表现出补体依赖的细胞毒性 (CDC) 活性，脱糖基化会大幅降低其活性，且在该实验中与杂交瘤来源的 AGS-PSCA 抗体效力相当^[1]。
Intact AGS-1C4D4 对体外培养的 HPAC 胰腺细胞无直接细胞毒活性^[1]。
AGS-1C4D4 与 PSCA 结合的 K_d 值为 2.0 × 10^-10 M，表现出与杂交瘤来源的 AGS-PSCA 抗体相似的结合亲和力^[1]。

  Flow Cytometry analysis of HepG2 cells with MK-4721 (HY-P991229, red) . Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then cells were stained with the primary antibody at 1/200 dilution for an hour at 4℃. Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa(HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
