NVP-BVB808 is a selective and ATP-competitive JAK2 inhibitor with an IC50 of 0.35 nM. NVP-BVB808 binds to JAK2’s ATP-binding site, stabilizes JAK2’s active conformation, increases JAK2 activation loop phosphorylation, and blocks downstream kinase function. NVP-BVB808 exhibits antiproliferative and pro-apoptosis effects, suppresses constitutive STAT5a phosphorylation. NVP-BVB808 can be used for the research of cancer, such as leukemia[1].

NVP-BVB808 (72 h) 可强效抑制携带 JAK2 突变的 CHRF-288-11、SET-2 和 UKE-1 细胞的增殖,72 小时后的 GI50 值分别为 0.15 μM、0.25 μM 和 0.22 μM;对 HEL 细胞的活性较弱 (GI50 = 2.13 μM);仅在较高浓度下才会抑制 BCR-ABL 阳性和 JAK2 野生型细胞系的增殖 (GI50 = 0.73-3.20 μM)[1]
NVP-BVB808 (72 h) 可诱导 CHRF-288-11、SET-2、HEL、Lama-87、KU-812、BV173、K562 和 KCL-22 细胞发生细胞凋亡,其中对 CHRF-288-11 细胞的活性最强 (IC50 = 0.3 μM),对其余细胞系的活性依次降低[1]
NVP-BVB808 (0-1 μM; 30 min) 可在低至 0.05 μM 的浓度下、作用 30 分钟后强效抑制 JAK2 突变型 SET-2 和 HEL 细胞中 STAT5a 的磷酸化,而对 BCR-ABL 阳性 K562 和 KCL-22 细胞仅表现出极微弱的作用[1]

Concentration: 0, 0.01, 0.05, 0.1, 0.5, 1 μM
Incubation Time: 30 min
Result: Suppressed constitutive STAT5a phosphorylation in SET-2 cells at 0.1 μM.
Suppressed constitutive STAT5a phosphorylation in HEL cells at ~0.5 μM.
Showed only weak to no inhibition of STAT5a phosphorylation in BCR-ABL-positive K562 and KCL-22 cells at tested concentrations.
