Paclitaxel liposome is a liposome-encapsulated form of Paclitaxel (HY-B0015). Paclitaxel is a naturally occurring antineoplastic agent and stabilizes tubulin polymerization. Paclitaxel liposome is characterized by reduced acute toxicity, prolonged circulating half-life of paclitaxel, and enhanced accumulation at tumor sites. Paclitaxel liposome can be used in research related to breast cancer and locally advanced esophageal squamous cell carcinoma[1][2].

Paclitaxel liposome (0.1-5000 ng/mL; 24-72 h) 对人乳腺癌 MCF-7 细胞具有强效、时间和浓度依赖性的细胞毒性,处理 72 h 后的 IC50 为 11.54 ng/mL[1]

Cell Cytotoxicity Assay[1]

Cell Line: human breast cancer MCF-7 cells
Concentration: 0.1-5000 ng/mL
Incubation Time: 24 h; 48 h; 72 h
Result: Showed concentration- and time-dependent inhibitory effects on MCF-7 cells.
Failed to achieve an IC50 after 24 h of treatment.
Reached an IC50 of 2095 ng/mL after 48 h of treatment.
Reached an IC50 of 11 ng/mL after 72 h of treatment.
体内研究
(In Vivo)

Paclitaxel liposome (5 mg/kg;静脉注射;每 2 天 1 次;连续给药 4 次) 可抑制裸鼠体内 MCF-7 乳腺癌肿瘤的生长,在治疗启动后第 34 天将平均肿瘤体积降至 1700 mm3,同时可支持裸鼠体重逐步增长[1]
Paclitaxel liposome (10 mg/kg;静脉注射;单剂量) 可适度延长紫杉醇在健康 ICR 小鼠体内的循环时间,其消除半衰期为 2.26 h,生物利用度较游离紫杉醇高 1.38 倍[1]
普通紫杉醇脂质体 (L-PTX)(80-200 mg/kg;静脉注射;单次给药) 可降低健康 ICR 小鼠的急性毒性,其 LD50 为 134.46 mg/kg[1]

Animal Model: BALB/c nude (female, 16-18 g, inoculated with MCF-7 human breast cancer cells)[1]
Dosage: 5 mg/kg (based on paclitaxel content)
Administration: i.v.; once every 2 days; 4 consecutive administrations
Result: Reached mean tumor volume of 1000 mm3 after 18 days of administration.
Reached mean tumor volume of 1700 mm3 after 34 days of administration.
Showed gradual increase in body weight throughout the study period.
