生物活性

RM2 is a GRPr antagonist. RM2 selectively binds to GRPr, blocks agonist-induced receptor internalization, and inhibits agonist-triggered intracellular calcium mobilization. RM2 can be used in studies related to prostate cancer[1].

(In Vitro)

RM2 以高亲和力结合人前列腺癌冰冻切片上的 GRP 受体，其 IC50 为 7.7 nmol/l[1]。

RM2 (1-10 μmol/l; 60 s) 可作为 PC-3 细胞中的 GRP 受体拮抗剂，在 1 μmol/l 或 10 μmol/l 浓度下对细胞内 Ca2+动员无固有活性，但在 10 μmol/l 浓度下可抑制蛙皮素诱导的 Ca2+动员[1]。

Concentration: 1000 nmol/l (single treatment); 1000 nmol/l (co-treated with 10 nmol/l bombesin)
Incubation Time: 30 min (37°C, single treatment); 30 min (37°C, co-treated with 10 nmol/l bombesin)
Result: Failed to stimulate GRP receptor internalization.
Prevented bombesin-induced GRP receptor internalization when co-treated with 10 nmol/l bombesin.

111In-RM2 (10 pmol；尾静脉注射) 在 PC-3 前列腺癌异种移植瘤中实现了高效、持续的 GRPr 特异性摄取，注射后 72 小时的瘤血比可达 4070[1]。
68Ga-RM2 (80 pmol；静脉注射) 在 PC-3 前列腺癌异种移植物中表现出 GRPr 特异性的高摄取，注射后 80 分钟时肿瘤与肾脏的比值可达 6.1[1]。
68Ga-RM2 (80 pmol；静脉注射) 在雄激素依赖性 LNCaP 前列腺癌异种移植物中实现 GRPr 介导的特异性摄取，注射后 120 分钟时肿瘤与血液的比值可达 23.4[1]。
