RPG-01-132 is a DENV capsid protein PROTAC degrader, with a DC50 of 2.4 μM. RPG-01-132 blocks viral assembly, eliminates viral particles, and interferes with the non-structural functions of capsid protein. RPG-01-132 exhibits significant antiviral activity against all four DENV serotypes and the ST148 (HY-121663)-resistant DENV2 mutant strain. RPG-01-132 is applicable to research related to dengue virus infection[1]. (Pink: Dengue Virus ligand (HY-121663); Blue: Cereblon E3 ligase ligand; Black: linker (HY-130481)).

RPG-01-132 (0.04-20 μM; 24 h) 可在野生型 Huh7.5 细胞中以浓度依赖、CRL4CRBN 依赖的方式降低感染性 DENV2 子代病毒的产量，其 EC50 为 0.47 μM；但在 CRBN 敲除的 Huh7.5 细胞中无显著活性[1]。
RPG-01-132 (30 μM; 24 h) 可强效抑制 Huh7.5 细胞中 DENV2 病毒粒子的组装，导致细胞内区室中几乎完全检测不到病毒粒子[1]。
RPG-01-132 (0.156-0.625 μM; 24 h) 可浓度依赖性地恢复 HEK293 细胞中由异位表达 DENV2 衣壳蛋白抑制的 IFN-β 启动子活性，表明其可破坏衣壳蛋白介导的干扰素拮抗作用[1]。
RPG-01-132 (0.04-20 μM; 24 h) 在 10 μM 浓度下对四种 DENV 血清型均表现出稳定的、依赖 CRL4CRBN 的抗病毒活性；相较于野生型 DENV2 16681，其对 ST148 耐药的 DENV2 16681 C-S34L 突变株仍保有相当的衣壳降解活性 (DC50 = 1.85 μM) 和抗病毒活性 (EC50 = 0.58 μM)[1]。
