STING agonist-48 is a potent STING agonist that exhibits STING-dependent activity in vitro (EC50 = 4.02 μM). STING agonist-48 prefers to bind with the transmembrane domain (TMD) over the cytosolic cyclic dinucleotide (CDN) domain. STING agonist-48 shows adjuvant efficacy, enhancing IgG and Th1/Th2 cytokine responses in humanized STING mice. STING agonist-48 can be used for the study of inflammation-related diseases[1].

STING agonist-48 (Compound 7k) (6.25-50 μM，24 小时) 可诱导 THF-ISRE IRF3 细胞中荧光素酶活性呈剂量依赖性增加，表明其具有强烈的 STING 激活作用；但在 THF-ISRE STING-/- 细胞中，不诱导荧光素酶活性，且该活性完全丧失；在 THF-ISRE TRIF/MAVS-/- 细胞中，仍能维持剂量依赖性的荧光素酶活性增加[1]。

Animal Model: Male and female C57BL/6 humanized STING (huSTING; 6-12 weeks) mice were intramuscularly immunized and boosted (at a two-week interval) with 50 μL of either detergent-split A/Victoria/210/2009 (H3N2, A/Vic)[1].
Dosage: 20 μg
Administration: I.m., 0 day, 14 days
Result: Serum IgG1, IgG2, and total IgG titers were significantly increased in mice.
Increased secretion of IFN-γ, IL-4, and IL-5 from spleen cells.
