SYTO9 (solution) 

SYTO9 (solution) is a green fluorescent nucleic acid dye (Ex=485 nm, Em=498 nm (DNA)/501 nm (RNA)). SYTO9 can penetrate the intact cell membrane of viable bacteria. When the cell membrane is damaged (cell death), its rate of entry into cells increases, and it competes with propidium iodide (PI) (HY-D0815), another membrane-impermeable dye, for nucleic acid binding sites. SYTO9 is widely used in fluorescence microscopy, flow cytometry, and fluorometric assays in microbiology for bacterial counting, viability assessment, and cell imaging^[1][2][3].

Solvent and concentration: DMSO: 5 mM

SYTO9 (0.5-1 μM; 5-30 min) 对 Bacillus cereus、Escherichia coli、Salmonella enterica 亚种肠炎血清型鼠伤寒菌株以及 Staphylococcus aureus 菌悬液的染色会产生与细胞浓度相关的种属特异性荧光强度；在测试范围内，1 mM EDTA、染色时长 (5-30 min) 或染料浓度 (0.5 μM vs. 1 μM) 均未产生显著影响^[1]。
SYTO9 (0.5-1 μM; 5-30 min) 在无细胞蛋白胨水中表现出：荧光强度随浓度 (0.5 μM vs. 1 μM) 升高出现微小但显著的提升，且 1 mM EDTA 或孵育时长 (5-30 min) 对其无显著影响^[1]。
SYTO9 (3.34 mM stock in DMSO; 15 min dark incubation post-staining; assessed at 0, 40, 80, 160, 240 min ethanol exposure time points) - PI 双染色可在Escherichia coli K-12 菌株 M72 (plucTet) 中区分活细胞、受损细胞和死细胞群体，但与 GFP-PI 染色相比，其群体分离效果较差，且经乙醇处理后 SYTO9 的绿色荧光强度最高可提升 3.9 倍^[2]。
SYTO9 (0.5 μM) 法高分辨率熔解曲线 (HRM) 分析可精准检测培养羊膜细胞或脐血细胞基因组 DNA 中的 β⁰-地中海贫血突变 (codons 17, 41/42, 71/72) 及 Hb-E，检测结果与胎儿血的 HPLC 分析结果完全一致^[3]。
