Tri-O-tolyl phosphate (Tri-O-cresyl phosphate; TOTP) is an aryl phosphate compounds commonly utilized as flame retardants and lubricant additives across various industrie. Tri-O-tolyl phosphate has neurotoxicity and reproductive toxicity, decreasing cell viability, inhibiting cell proliferation, triggering cell cycle arrest, induceing apoptosis, causing mitochondrial dysfunction, disrupts calcium homeostasis[1][2].

Tri-O-tolyl phosphate (0-200 ppm; 48 h) 以浓度依赖方式降低 HTR-8/SVneo 和 JEG-3 滋养层细胞的活力,在 200 ppm 处理 48 h 时,HTR-8/SVneo 和 JEG-3 细胞的活力分别降至 0.28 倍和 0.05 倍[1]

Cell Line: HTR-8/SVneo human trophoblast cells, JEG-3 human choriocarcinoma trophoblastic cells
Concentration: 0, 5, 10, 20, 50, 100, 200 ppm
Incubation Time: 48 h
Result: Reduced relative cell viability to 0.69-fold at 5 ppm, 0.27-fold at 10 ppm, and 0.28-fold at 200 ppm in HTR-8/SVneo cells.
Reduced relative cell viability to 0.57-fold at 5 ppm, 0.43-fold at 10 ppm, and 0.05-fold at 200 ppm in JEG-3 cells.

Cell Cycle Analysis[1]

Cell Line: HTR-8/SVneo cells, JEG-3 cells
Concentration: 1, 2, 5 ppm
Incubation Time: 48 h
Result: Increased the percentage of HTR-8/SVneo cells in subG1 phase from 12.05% to 18.12%, in S phase from 11.91% to 25.96%, and in G2/M phase from 18.61% to 30.65%, while decreasing the percentage in G1 phase from 57.43% to 25.27% at 5 ppm.
Increased the percentage of JEG-3 cells in subG1 phase from 27.90% to 67.29%, while decreasing the percentage in G1 phase from 50.46% to 7.01% at 5 ppm.

Western Blot Analysis[1]

Cell Line: HTR-8/SVneo cells, JEG-3 cells
Concentration: 1, 2, 5 ppm
Incubation Time: 3 h
Result: Upregulated p-ERK1/2 by 1.41-fold, p-JNK by 2.13-fold, and p-AKT by 5.97-fold, while downregulating p-S6 by 0.61-fold in HTR-8/SVneo cells at 5 ppm.
Upregulated p-ERK1/2 by 1.67-fold, p-JNK by 2.24-fold, and p-AKT by 1.86-fold, while downregulating p-S6 by 0.49-fold in JEG-3 cells at 5 ppm.
