BXY-14 is a TLR2 agonist and vaccine adjuvant. BXY-14 significantly downregulates the expression of intratumoral PD-L1 in mouse models. BXY-14 acts as a vaccine adjuvant to induce antibody responses. BXY-14 exhibits synergistic efficacy when combined with the anti-PD-L1 monoclonal antibody Atezolizumab (HY-P9904) in mouse models of melanoma, and prolongs overall survival. BXY-14 is applicable to research related to melanoma[1].

Animal Model: C57BL/6 (female, 6-8 weeks old, ~20 g)[1]
Dosage: 10 mg/kg; 20 mg/kg
Administration: s.c.; on days 0, 14, and 28
Result: Induced significantly stronger total IgG, IgG1, and IgG2c anti-OVA antibody titers than positive control adjuvant Diprovocim on day 42, with a marked Th1-skewed immune bias (higher Th1/Th2 index) at 10 mg/kg.
Induced robust total IgG, IgG1, and IgG2c anti-OVA antibody titers, with no significant body weight changes observed over 10 days post-immunization, indicating good tolerance at 20 mg/kg.
Animal Model: C57BL/6 (female, 6-8 weeks old, ~20 g, subcutaneously injected with 2×105 B16-OVA melanoma cells on day 42 post-initial immunization)[1]
Dosage: 10 mg/kg (in combination with anti-PD-L1); 20 mg/kg (monotherapy)
Administration: s.c.; on days 0, 14, and 28
Result: Achieved stronger tumor growth inhibition than Diprovocim plus anti-PD-L1, prolonged overall survival to 100% at day 50, significantly increased tumor cell apoptosis (higher mean fluorescence intensity of TUNEL staining), and markedly downregulated intratumoral PD-L1 expression (lower integrated optical density of PD-L1 staining) at 10 mg/kg in combination with anti-PD-L1.
Potently suppressed tumor growth, prolonged survival, and downregulated intratumoral PD-L1 expression at 20 mg/kg as monotherapy.
